Enhancement of heme synthesis in quail embro yolk sac cells by highly polar compounds.

چکیده
Following our previous paper, we exammed the effects on the heme synthesis in quail yolk sac cells of polar compounds, other than dimethyl sulfoxide. that induce erythroid cell differentiation in cultured Friend leukemia cells. Dissociated yolk sac cells from quail embryos at the definitive primitive streak stage were cultured in a gyratory shaker in a medium containing dimethyl sulfoxide or other polar compounds. The single aggregates thus formed were further cultured in an egg agar medium containing Fe. An increase in heme synthesis was observed in the explants incubated in dimethyl sulfoxide, N,N-dimethylacetamide, 1 -methyl-2-piperidone. N-methylacetamide. or triethylene glycol, but not in those incubated in acetamide, propionamide. or N.N-dimethylformamide. A weak increase was observed in those incubated in hexamethylene bisacetamide. The hemoglobin and DNA contents were chemically determined in the pooled explants treated with N.N-dimethylacetamide to give an example of a polar compound. There was an obvious increase in hemoglobin content in the treated explants. The DNA content of the treated explants was also higher than in the control. The cellular permeability for Fe was not affected by N.N dimethylacetamide.

متن کاملAre developmental hemoglobin changes related to the origin of stem cells and site of erythropoiesis?
HemoglobIn ontogenic changes have been studied In chimeras composed of a quaIl embryo grafted on a chIck yolk sac. From the blood of such chlmeras, quaIl erythrocytes are separated from chIck by means of a specific Immune hemolytlc technIque, thus yielding two populations of red cells, one derIving from yolk sac stem cells and the other from intraembryonlc stem cells. Both populations comprIse ...
متن کاملStimulation of haem synthesis by erythropoietin in mouse yolk-sac-stage embryonic cells.
Cultures of disaggregated cells from mouse embryos at the stage of yolk-sac erythropoiesis have been used to test sensitivity of primary erythroid cells to erythropoietin, the hormone which controls adult red cell production. Synthesis of haem was stimulated by 89% in cells of 8-day embryos, 23 % in cells of 9-day embryos and by 19% in peripheral nucleated blood cells of 12-day embryos. Differe...
متن کاملSynthesis of embryonic hemoglobins during erythroid cell development in fetal mice.
Three embryonic hemoglobins are formed in erythroid cells derived from yolk sac blood islands in fetal mice of the C57BL/6J strain. These cells provide a suitable model to study the rates of synthesis of specific proteins during the differentiation of cells. Erythroid cells differentiate initially in yolk sac blood islands from the 8th to the 10th day of gestation. (Fetal mice have a ZOto Zl-da...
متن کاملExtraembryonic Origin of Circulating Endothelial Cells
Circulating endothelial cells (CEC) are contained in the bone marrow and peripheral blood of adult humans and participate to the revascularization of ischemic tissues. These cells represent attractive targets for cell or gene therapy aimed at improving ischemic revascularization or inhibition of tumor angiogenesis. The embryonic origin of CEC has not been addressed previously. Here we use quail...
